Modeling of the Singlet Oxygen Distribution in Photofrin- Photodynamic Therapy of the Plural Cavity

چکیده
Photofrin-mediated photodynamic therapy (PDT) is used after surgical resection at the University of Pennsylvania to treat the microscopic disease for malignant pleural mesothelioma and to increase survival rate. When Photofrin is exposed to laser light at 630 nm in well-oxygenated tissue, it produces reacted singlet oxygen ([O2]rx) that kills cancer cells. As [O2]rx is imperative to PDT treatment efficacy, in this study we use COMSOL Multiphysics software to simulate the distribution of [O2]rx on every point of the plural cavity surface that is being treated. The geometry of the pleural cavity that is used in this study was obtained during the surgery using an infrared camera–based navigation system (NDI). The initial Photofrin concentration inside the pleural cavity and the light fluence information used for the simulation of [O2]rx were measured during the treatment.

Photosensitization of coronene–purine hybrids for photodynamic therapy
Photosensitization properties of coronene-purine (Cor-P) hybrids for photodynamic therapy (PDT) have been investigated in this work. Eight hybrid Cor-P models have been designed by the additional of adenine (A) and guanine (G) nucleobase to Cor species. The evaluated absorption and emission energies indicated that the singular models are not good at all for PDT process whereas their hybrid mode...
متن کاملErratum: Macroscopic singlet oxygen modeling for dosimetry of Photofrin-mediated photodynamic therapy: an in-vivo study.
Although photodynamic therapy (PDT) is an established modality for cancer treatment, current dosimetric quantities, such as light fluence and PDT dose, do not account for the differences in PDT oxygen consumption for different fluence rates ( ? ). A macroscopic model was adopted to evaluate using calculated reacted singlet oxygen concentration ( [ O 2 1 ] rx ) to predict Photofrin-PDT outcome i...
متن کاملA Comparison of Singlet Oxygen Explicit Dosimetry (SOED) and Singlet Oxygen Luminescence Dosimetry (SOLD) for Photofrin-Mediated Photodynamic Therapy
Accurate photodynamic therapy (PDT) dosimetry is critical for the use of PDT in the treatment of malignant and nonmalignant localized diseases. A singlet oxygen explicit dosimetry (SOED) model has been developed for in vivo purposes. It involves the measurement of the key components in PDT-light fluence (rate), photosensitizer concentration, and ground-state oxygen concentration ([³O₂])-to calc...
متن کاملSensitivity of Candida albicans germ tubes and biofilms to photofrin-mediated phototoxicity.
Treatment of mucocutaneous and cutaneous Candida albicans infections with photosensitizing agents and light, termed photodynamic therapy (PDT), offers an alternative to conventional treatments. Initial studies using the clinically approved photosensitizer Photofrin demonstrated the susceptibility of C. albicans to its photodynamic effects. In the present study, we have further refined parameter...
متن کاملPhotofrin photodynamic therapy can significantly deplete or preserve oxygenation in human basal cell carcinomas during treatment, depending on fluence rate.
At high fluence rates in animal models, photodynamic therapy (PDT) can photochemically deplete ambient tumor oxygen through the generation of singlet oxygen, causing acute hypoxia and limiting treatment effectiveness. We report that standard clinical treatment conditions (1 mg/kg Photofrin, light at 630 nm and 150 mW/cm2), which are highly effective for treating human basal cell carcinomas, sig...
